AMINO ACID DERIVATIVES & UNCA’S

An example of this situation is the production of Fmoc-amino acids. It is not unusual for Fmoc amino acids to contain non-negligible amounts of Fmoc-beta-alanine and Fmoc-beta-alanine-AA. Impurities in the final peptide arising from these contaminants are often extremely difficult to remove, resulting in the lowering of the yield and increasing the cost. Non-Natural Amino acid derivatives:

One of the major challenges of using peptides as therapeutics is their susceptibility to enzymatic proteolysis. Replacement of certain amino acid residues by small organic moieties (building blocks) or unnatural amino acids often leads to peptides with enhanced resistance to proteolysis. NCAs and UNCAs:

Amino acid N-carboxy anhydrides (NCAs) and Urethane protected NCAs (UNCAs) are valuable tools for the synthesis of polyamino acids (NCAs) and peptides (UNCAs). These molecules are activated and protected (in the case of UNCAs), and react with nucleophiles to provide amino acid derivatives with CO2 as the only co-product.
